Material and method: we carried out a descriptive, cross-sectional, prospective study in 70 mothers of premature babies in the unit. intermediate care in the Hospital. Mothers without debilitating or immunosuppressive diseases, with a newborn without severe complications during pregnancy, and who had voluntarily agreed to participate in the project with the informed consent form were included. The technique used was the survey, through the questionnaire modified by Carranza & Palma (2017) and validated for this study called “Knowledge Questionnaire of Mothers of Premature Babies about Breastfeeding” – CCOMLA, which consists of 18 questions. Results: Of the total, 48.6% corresponded to the age group of 30-39 years, they were mostly married (35.7%), with a higher education degree (45.7%). Also, 38 (54.3%) mothers had a dependent occupation and 50 (71.4%) mothers had a previous child. The gestational age of the most frequent was 32 weeks (50%), with a hospitalization time of >15 days (68.6%). Of the total, 57.1% (40 mothers) had medium knowledge and 38.6% (27 mothers) had a high level of knowledge. Conclusion: There is a moderate level of knowledge of mothers about breastfeeding in premature neonates in the intermediate care unit at the Edgardo Rebagliati Martins Hospital, 2020.
