The objective of this research is to evaluate the relationship between knowledge and compliance with the Manual of Good Pharmaceutical Office Practices (MBPOF) among pharmaceutical personnel at the Mifarma pharmacy chain in the Surco district, 2022. Methodology: This is a non-experimental, correlational, observational, and cross-sectional study. The technique used was a survey, and the instrument was a questionnaire consisting of 34 dichotomous and multiple-choice questions based on the Likert scale. The study included 256 participants (pharmaceutical and technical staff) to identify deficiencies and help improve dispensing, distribution, and medication control conditions. To test both general and specific hypotheses, Cramer’s V statistic was used to observe correlations between variables, as well as the binomial test for hypotheses with dichotomous variables. Results: It was found that 96.9% of participants were knowledgeable about the MBPOF. Moreover, a relationship was identified between knowledge and compliance with this manual, with Cramer’s V showing p = 0.00 < 0.05. Additionally, 97.3% of participants were familiar with Good Storage Practices (GSP), 95.3% with Good Dispensing Practices (GDP), and 89.8% with Good Pharmacovigilance Practices (GPP). Regarding compliance, 17.3% always adhered to GSP, 43.8% to GDP, and 1.6% to GPP. Conclusion: There is a relationship between knowledge and compliance with the Manual of Good Pharmaceutical Office Practices among pharmaceutical personnel at Mifarma pharmacies in the Surco district, 2022.
