The objective was to validate the suitability of the microbiological method for microbial count and specific microorganisms in coated tablets of Sulfamethoxazole + Trimethoprim, applying both the direct inoculation and membrane filtration methods in accordance with the official standards of the United States Pharmacopeia (USP 42). The results demonstrated that the direct inoculation method, using neutralizing broth medium, was the most appropriate, as it produced more accurate and precise microbial counts across the three test batches. The results complied with the established parameters, meeting the requirements for suitability, growth promotion, and strain standardization, thus validating the microbiological test. On the other hand, the membrane filtration method yielded limited recovery due to the amount of supernatant generated by the sample when diluted with the culture medium, making it unsuitable for solid products. In conclusion, the direct inoculation method proved suitable for microbial count and specific microorganism analyses, while the membrane filtration method was not appropriate for microorganism recovery in coated tablets.
