Publicación Acceso abierto Concordancia entre el método en microplacas y cromatografía de alta performace para la medición de Zinc protoporfirina en sangre humana, Lima, Noviembre 2025(Universidad Privada Norbert Wiener, 2025-09-21) Cabanillas Espinoza, Carmen Elizabeth; Huamán Cárdenas, Víctor RaúlEl presente estudio tiene por finalidad determinar la concordancia entre el método en microplacas y la cromatografía líquida de alta performance (HPLC) para la determinación de zinc protoporfirina (ZPP) en sangre humana. La ZPP es un biomarcador de efecto útil para el diagnóstico de la intoxicación por plomo, problemática que se da en poblaciones expuestas a actividades mineras, como es el caso de Perú. El método gold estándar HPLC requiere de equipamiento costoso, es por ello que, en este estudio se propone la validación de un método alternativo, más sencillo y más accesible de realizar usando microplacas, con extracción alcohólica y lectura fluorométrica. Se analizarán 80 muestras de sangre venosa anticoagulada con EDTA, protegidas de la luz, recolectadas en un hospital de Lima durante el mes de noviembre de 2025. El presente estudio sigue un diseño observacional, transversal y prospectivo, con un enfoque cuantitativo. Se utilizarán gráficos de Bland-Altman y el Coeficiente de Correlación Intraclase (CCI) como pruebas para determinar la concordancia entre ambos métodos; se espera que el método en microplacas sea clínicamente intercambiable con el HPLC, permitiendo así aumentar el acceso a pruebas de biomarcadores, mejorando así el seguimiento de las personas expuestas a contaminación por plomo.Publicación Acceso abierto Fracciones lábil y estable de HBA1C basal y su relacion con la glicemia basal y post tolerancia oral en pacientes atendidos en el laboratorio Suiza Lab, Lima 2017(Universidad Privada Norbert Wiener, 1905-07-09) Arrarte Castro, Corina Doris; Delgado Ortiz, Sara; Sandoval Vegas, Miguel HernánSe realizó una selección de 1455 pacientes a los cuales se le han solicitado las pruebas de HbA1c, glucosa basal y tolerancia a la glucosa en julio a setiembre del 2017.El presente estudio es de tipo cuantitativo, básico, retrospectivo, transversal, exploratorio, el diseño de investigación es sin intervención. Resultados obtenidos se analizaron estadísticamente descriptiva mediante el promedio, la mediana, desviación estándar de las pruebas de hemoglobina glicosilada fracción lábil y estable, glucosa basal y tolerancia a la glucosa. Con respecto a la HbA1c, LA1C, SA1C, Glucosa basal y T 120 min según sexo no existe diferencia significativa en los pacientes. The samples were dried and pulverized, followed by defatting with 500 mL of petroleum ether using the Soxhlet method. Hydrolysis was carried out with 6N HCl. The identification of essential amino acids in Vigna unguiculata and Phaseolus vulgaris was performed using High-Performance Liquid Chromatography (HPLC). The results showed that Vigna unguiculata contained valine (1.02 mg/100 mg), glycine (8.75 mg/100 mg), leucine (1.01 mg/100 mg), lysine (0.64 mg/100 mg), and methionine (1.05 mg/100 mg), while Phaseolus vulgaris contained valine (1.79 mg/100 mg), glycine (5.49 mg/100 mg), tryptophan (1.33 mg/100 mg), leucine (1.08 mg/100 mg), and lysine (1.31 mg/100 mg). Linearity was demonstrated by obtaining a correlation coefficient of r = 0.998 for the system and 0.9996 for the method, values that exceed the minimum permissible limit of 0.995. Accuracy was evaluated using the Student’s t-test, yielding a texp value of 1.1444, which is lower than the tabulated value of 2.306; therefore, no significant difference was found between the mean recovery and the amount added, indicating that the percentage recovery is close to 100%. Precision was confirmed by the relative standard deviation (RSD) values obtained, which were 0.19% for repeatability and 0.52% for intermediate precision, both well within the maximum allowed RSD of 2.0%. Specificity was also established, as the chromatographic peaks obtained showed similar retention times for both the sample and the standard, confirming the specificity of the method. Furthermore, analysis of the standard and placebo demonstrated that no excipients interfered with the active ingredient peak, nor was the presence of degradation products of Levocetirizine Dihydrochloride detected under forced degradation conditions. These results confirm that the method is selective.Publicación Acceso abierto Validación del método analítico por cromatografía líquida de alta performance para la cuantificación de nitazoxanida 100mg/ 5ml polvo para suspensión oral(Universidad Privada Norbert Wiener, 2015) Gomez Leon, Carmen Rosa; Ramos Silva, Hesica; Villanueva Vílchez, Hugo GilbertoIn this research work, the validation of an analytical method for Nitazoxanide 100 mg/5 mL powder for oral suspension was developed using the High-Performance Liquid Chromatography (HPLC) technique. The validation of the method is an indispensable requirement for compliance with Good Laboratory Practices, since it provides reliability to the analytical results obtained in an analytical laboratory, in order to ensure that a medicine complies with the quality parameters established by USP 37. Once the working parameters were defined, the validation protocol of the developed method was carried out, which includes the experimental design and statistical procedures. The quality parameters employed in this validation are: Linearity, which measures the ability of the analytical method to provide results that are directly proportional to the concentration of the analyte, obtaining a correlation coefficient of r = 0.99960. Accuracy, which measures the closeness of the results obtained by this method, with an experimental t (0.633) that is lower than the tabulated t (2.306). Precision, which expresses the degree of agreement or dispersion of the results; the RSD values obtained were 0.697% for method repeatability and 0.913% for intermediate precision. Selectivity, which determines the ability of the method to unequivocally measure the Nitazoxanide content in the presence of other chemical substances that may be present in the sample or degradation products; in the chromatograms of Nitazoxanide, similar retention times were obtained for the sample and the standard. It was demonstrated that the validated analytical method is reproducible, reliable, and can be employed in routine analyses.
