Actividad antimicrobiana del aceite esencial de Origanum vulgare L. “orégano peruano” frente a Staphylococcus aureus

Abstract

The objective of this research was to confirm the antimicrobial activity of the essential oil of Origanum vulgare L., "Peruvian Oregano," against Staphylococcus aureus. The essential oil was obtained by steam distillation and purified with anhydrous sodium sulfate. The analysis of the main phytochemical components of the oil was performed using organoleptic and instrumental methods. The antimicrobial activity was assayed against Staphylococcus aureus ATCC 6538 using the disk diffusion method (Kirby-Bauer), with Gentamicin 10 µg (OXOID brand) as the positive control and dimethyl sulfoxide as the negative control. Results: The essential oil had a good yield of 2.02%, distilled in 60 minutes, with a density of 0.9132 g/mL and a refractive index of 1.475. The main components were carvacrol and thymol. The essential oil of Origanum vulgare L. showed antimicrobial activity against Staphylococcus aureus ATCC 6538, with inhibition halos of 11.47,13.40,16.60, and 19.65 mm at concentrations of 25,50,75, and 100 µg/mL, respectively. The gentamicin control resulted in an inhibition halo of 30.14 mm. Conclusion: The highest antimicrobial activity of the essential oil of Origanum vulgare L. "Peruvian Oregano" against Staphylococcus aureus was confirmed at 100 µg/mL.

El objetivo del presente trabajo de investigación fue comprobar la actividad antimicrobiana del aceite esencial de Origanum vulgare L.“Orégano peruano” frente a Staphylococcus aureus”. El aceite esencial se obtuvo por el método de destilación de arrastre con vapor de agua, purificándose con sulfato de sodio anhidro. El análisis de los principales componentes fitoquímicos del aceite se realizó por métodos organolépticos e instrumentales. La actividad antimicrobiana se ensayó en cepas de Staphylococcus aureus ATCC 6538 por el método de difusión en disco (Kirby-Bauer) en Placas de Petri, el control positivo fue con discos de Gentamicina 10 µg (marca OXOID) y el control negativo con dimetilsulfóxido. Los resultados obtenidos fueron los siguientes: el aceite esencial tiene un buen rendimiento siendo 2,02 %, destilado en un tiempo de 60 minutos, densidad de 0,9132 g/mL, índice de refracción de 1,475, y siendo los principales componentes el carvacrol y timol, el aceite esencial de Origanum vulgare L. “Orégano peruano” tiene actividad antimicrobiana frente a Staphylococcus aureus ATCC 6538 , los halos de inhibición fueron 11,47, 13,40, 16,60 y 19,65 mm , a las concentraciones de 25, 50, 75 y 100 µg/mL respectivamente. Para la gentamicina resultó un halo de inhibición de 30,14 mm. Se concluyó a 100 µg/mL la mayor actividad antimicrobiana el aceite esencial de Origanum vulgare L. “Orégano peruano” frente a Staphylococcus aureus.
