"Chuillur" en ratones(Universidad Privada Norbert Wiener, 2018-09-18) Picho Hurtado, Katheryn Elizabeth; Chávez Flores, Juana ElviraThe investigation was carried out on the plant species Vallea stipularis L.f. “Chuillur,” located in the district of Tamburco-Abancay-Apurímac, traditionally used by the local population as medicine for the treatment of scurvy, as a cicatrizant, for gastritis, rheumatism, drastic purgative, and analgesic. Objective: To verify the analgesic activity of the ethanolic extract of the root of Vallea stipularis L.f. “Chuillur” in mice. Methods: The method of abdominal writhing induced by 0.8% acetic acid was employed. Forty-nine albino mice of the species Mus musculus strain Balb/C53/CNPB, of both sexes, were randomly distributed into seven groups. Ethanolic extracts at doses of 50, 100, and 200 mg/kg were used, as well as the standards Paracetamol Q.P. 300 mg/kg and Tramadol Hydrochloride Q.P. 40 mg/kg. These were administered orally, and 30 minutes later, 0.8% acetic acid was administered intraperitoneally. The animals were immediately observed for 20 minutes. Results: All groups showed a significant reduction (p value = 0.000), obtaining differentiated analgesic effects. The maximum effect was achieved with the 200 mg/kg extract, with 70% inhibition, surpassing Paracetamol (55%). In the present work, the analgesic activity of the ethanolic extract of the shells of the seeds of Persea americana Mill “Strong Avocado” was verified in albino mice. With the method of Koster et al Modified of writhings induced by 0.6% glacial acetic acid used as a writhing stimulant, the concentrations evaluated were 50, 100 and 200 mg / kg, of extract of the shells of the seeds Persea americana Mill “Strong Avocado”, compared with Tramadol 40 mg / kg and acetylsalicylic acid 200 mg / kg. Obtaining a percentage of reduction in contortions with acetylsalicylic acid 200 mg/kg 88%, Tramadol 40 mg/kg 94%, Persea americana Mill 100 mg/kg 88%, Persea americana Mill 200 mg/kg 94%. The root was macerated for 7 days in 70% ethanol. Solubility and qualitative analyses were performed, and the extract was determined to be soluble in polar solvents and to contain flavonoids and alkaloids. To determine the analgesic activity, the 0.8% glacial acetic acid-induced abdominal writhing model was used on 49 Balbin/C53/CNPB strain mice. The groups were a negative control (distilled water), a positive control (0.8% acetic acid), the ethanolic extract at 25, 50, and 100 mg/kg, and comparative standards of paracetamol (300 mg/kg) and tramadol (40 mg/kg). To determine the anti-inflammatory effect, the subplantar edema method was used, with dexamethasone (4 mg/kg) and diclofenac (50 mg/kg) as standard drugs. The evaluation of the extract's analgesic activity showed an effective dose of 100 mg/kg with 69% writhing inhibition. The anti-inflammatory activity was effective at a dose of 25 mg/kg, with an inhibition percentage evolving from 56% to 99% over a six-hour period.Publicación Acceso abierto Actividad analgésica y antiinflamatoria del extracto etanólico de las flores de Bidens andicola H.B.K. For the analgesic activity, the modified Koster et al. model was employed, and the study groups were: negative control (distilled water), positive control (0.8% glacial acetic acid), ethanolic extract of Bidens andicola H.B.K. “quiquo” flowers at 50, 100, and 200 mg/kg, paracetamol 300 mg/kg, and tramadol 40 mg/kg. For the anti-inflammatory activity, the plantar edema method with 1% albumin was used, and the groups were: negative control (distilled water), ibuprofen 400 mg/kg, dexamethasone 1 mg/kg, and the ethanolic extract at 200, 400, and 600 mg/kg. The results show that the ethanolic extract of Bidens andicola H.B.K. “quiquo” flowers at 100 mg/kg (65%) and 200 mg/kg (70%) exhibits analgesic activity, lower than tramadol (91%) but comparable to paracetamol (81%). Regarding the anti-inflammatory activity, the ethanolic extract at 200, 400, and 600 mg/kg demonstrated efficacy, with the 600 mg/kg dose showing the greatest anti-inflammatory effect (100%), similar to dexamethasone and ibuprofen. It was confirmed that the ethanolic extract of Bidens andicola H.B.K. “quiquo” flowers possesses both analgesic and anti-inflammatory activities.Publicación Acceso abierto Actividad analgésica y antiinflamatoria del extracto hidroalcohólico de las hojas de Passiflora edulis Sims "maracuyá"(Universidad Privada Norbert Wiener, 2019-03-06) Jalixto Alata, Sonia Carola; Salas Gutierrez, Cristina Angélica; Chávez Flores, Juana ElviraThe species Passiflora edulis Sims, or "maracuyá," is a plant that grows in northern Peru, specifically in the Lima Department, Barranca Province, Supe district, in the Venado Muerto Ravine. This study aimed to evaluate the analgesic and anti-inflammatory activity of the hydroalcoholic extract from the leaves of Passiflora edulis Sims. Methodology: A total of 56 Holtzmann strain rats of different sexes were used for the evaluation of inflammation. The rats were divided into 8 groups of seven animals. Inflammation was induced using the 0.1 mL albumin-induced plantar edema method, and the plantar volume was measured with a plethysmometer every hour for 6 hours. For the analgesic activity evaluation, the 0.8% acetic acid-induced abdominal writhing method was employed. A total of 49 Mus musculus strain mice of different sexes were divided into 7 groups of seven animals. The number of abdominal writhes was individually quantified over a 20-minute period. Results and Conclusion: The results demonstrated anti-inflammatory activity, as the 800 mg/kg hydroalcoholic extract inhibited the development of plantar edema with an effect of 100% within one hour of application, comparable to 4 mg of dexamethasone. The study also showed the analgesic activity of the 400 mg/kg hydroalcoholic extract, which achieved an effect of 70%.Publicación Acceso abierto Actividad analgésica y antiinflamatoria del extracto hidroalcólico de hojas y tallos de Leucheria daucifolia (D. Don.) Crisci “Churoq wasin”. For the analgesic activity, the model of abdominal writhing induced by 0.8% acetic acid was employed; 49 mice were used, and the evaluated groups were: negative control (distilled water), positive control (0.8% acetic acid), hydroalcoholic extract at 50, 100 and 200 mg/kg, compared with paracetamol 300 mg/kg and tramadol 40 mg/kg. For the anti-inflammatory effect, the auricular edema method was used with an irritant agent (6% xylene diluted in acetone) applied to the mouse auricle. A total of 49 albino mice were used, and the study groups were: vaseline cream, creams prepared with the hydroalcoholic extract at 0.5, 1 and 2%, compared with diclofenac 1% cream and hydrocortisone 1% cream. The results show that the hydroalcoholic extract of leaves and stems of Leucheria daucifolia (D. Don.) Crisci “Churoq wasin” at a dose of 100 mg/kg presents analgesic activity with an inhibition percentage of writhing of 62%, not surpassing tramadol (86%) but comparable to paracetamol (76%). Regarding anti-inflammatory activity, the cream with 2% extract presented an inhibition percentage of inflammation of 48.7%, which is comparable to hydrocortisone 1% and diclofenac 1%. The objective of this study was to determine the anxiolytic and antidepressant activity of the essential oil from the seeds of Foeniculum vulgare Mill., 'fennel,' in albino mice. Methodology: The methodology used was experimental, explanatory, quantitative, and applied. The seeds were collected in the province of Huarochirí (Matucana). The essential oil (EO) was extracted by hydrodistillation. The anxiolytic activity was evaluated using the marble burying method in 40 mice at oral doses of 2.5, 5, and 10 mL/kg of EO, compared to diazepam at 10 mg/kg. The antidepressant activity was evaluated using the forced swim test in 40 mice at oral doses of 2.5, 5, and 10 mL/kg of EO, compared to fluoxetine at 20 mg/kg. Results: The essential oil of Foeniculum vulgare showed antidepressant activity at the first two doses and anxiolytic activity at all three doses. Conclusion: The essential oil demonstrated antidepressant activity at doses of 2.5, 5, and 10 mL/kg and showed activity comparable to diazepam.Publicación Acceso abierto Actividad anti-tirosinasa y efecto fotoprotector del extracto etanólico del tegumento de dos variedades de Phaseolus vulgaris L., ñuña roja y negra(Universidad Privada Norbert Wiener, 2020-12-01) Mera Santa Cruz, Ermitanio; Papuico Sanchez, Liliana Eulalia; Chávez Flores, Juana ElviraThe objective of this study was to determine the anti-tyrosinase activity and evaluate the photoprotective effect of the ethanolic extract from the tegument of two varieties of Phaseolus vulgaris L., red and black 'ñuña'. Methodology: Anti-tyrosinase activity was determined with the thin-layer chromatographic method using a 0.05% Kojic acid standard. For the photoprotective effect, 35 female Mus musculus Balb/C53 strain mice, seven weeks old, were divided into five experimental groups. UV-C radiation was applied to their backs for 30 minutes once a day for one month. Acute photodamage was evaluated using the Draize scale, and photoaging was evaluated using the Glogau scale. Results: Secondary metabolites such as polyphenols and flavonoids were found. The extract was not toxic in mice via oral and dermal administration at a limit dose of 5000 mg/kg. It has anti-tyrosinase activity, and when applied in a 5% gel, the red variety showed a greater effect. It also has a photoprotective effect in mice subjected to UVC radiation. Conclusion: The ethanolic extract from the tegument of the black 'ñuña' at 5% in gel has a photoprotective effect.Publicación Acceso abierto Actividad antibacteriana de los extractos de acetato de etilo y acetona del zumo de Punica granatum L. (“pomegranate”) has been used in traditional medicine since ancient times for the treatment of various diseases. Several studies have demonstrated its antioxidant, anti-inflammatory, antiparasitic, and antifungal properties, mainly from hydroalcoholic extracts of the peel. However, few studies have investigated pomegranate juice extracts using acetone and ethyl acetate. The aim of this research was to evaluate the antibacterial activity of acetone and ethyl acetate extracts from pomegranate juice. Antibacterial activity was assessed using the disk diffusion method. Four concentrations (200 mg/mL, 300 mg/mL, 400 mg/mL, and 500 mg/mL) of each extract were tested against Staphylococcus aureus ATCC 25923, Escherichia coli ATCC 25922, and Pseudomonas aeruginosa ATCC 27853. Inhibition zones were qualitatively determined according to the Durafford scale for in vitro inhibitory effects in phytotherapy and CLSI antimicrobial standards. The results indicated that the acetone extract exhibited antibacterial activity against S. aureus and E. coli. Furthermore, a comparative analysis revealed that microbial inhibition was inversely proportional to the quantification of ellagic acid. Kuntze ex Thell, “Canchalagua,” from the city of Ayacucho, was obtained at the Wholesale Market “La Parada,” district of La Victoria – Lima, and was subsequently taxonomically classified at the Natural History Museum of the Universidad de San Marcos. The sample was cleaned and dried at the Research Center of the Universidad Norbert Wiener, where the parts of the plant—stem, root, leaves, and flowers—were separated. Hydroalcoholic extracts of each part were prepared, and the phytoconstituents present were identified, such as flavonoids, reducing sugars, phenolic compounds, and carbohydrates. For the microbiological test, the Disk Diffusion method was used with blood agar medium and the Propionibacterium acnes strain (ATCC® 11827). Standard antibiotics Q.P.: Doxycycline, Levofloxacin, Azithromycin, and Penicillin were employed. The Objective of this research was to determine the antibacterial activity against Cutibacterium acnes and the antioxidant activity of the hydroalcoholic extracts from the leaves, flowers, stem, and root of Hypericum silenoides Juss, 'Canchalagua.' The Method employed consisted of preparing the hydroalcoholic extracts of the leaves, flowers, stem, and root of Hypericum silenoides Juss, followed by a solubility test and qualitative analysis to identify the metabolites. To determine the antibacterial activity against Cutibacterium acnes (ATCC 6919), the disk diffusion method was performed. For antioxidant activity, the 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical method was used for evaluation. Results: The microbiological assay showed that the positive control group presented a larger diameter of the inhibition halo compared to the plant species extracts studied. The hydroalcoholic extract of the flowers showed greater antibacterial activity compared to the extracts of the leaves, stem, and root. The hydroalcoholic extract of the leaves of Hypericum silenoides Juss, 'Canchalagua,' presented the highest antioxidant activity with an IC50 of 54.53 µg/mL, followed by the hydroalcoholic extract of the flowers with an IC50 of 57.05 µg/mL, compared to the control (Trolox) with a p < 0.05. Conclusion: The hydroalcoholic extracts of the leaves, flowers, stem, and root have antibacterial activity against Cutibacterium acnes and antioxidant activity.Publicación Acceso abierto Actividad anticonvulsivante del extracto hidroalcoholico de raíz de Valeriana pinnatifida R. & P. "Valeriana"(Universidad Privada Norbert Wiener, 2018-11-24) Anco Maximiliano, Yanet Soleda; Chávez Fernández, Ana MaríaValeriana pinnatifida is a native plant of the Valerianaceae family, endemic to Peru, which grows on rocky slopes in the Andes. Traditionally, it has been used as a sedative, hypnotic, and tranquilizer. This study was initiated to analyze the metabolites present in the plant, determine its acute toxicity, and evaluate its anticonvulsant effect. The research was quasi-experimental. For the phytochemical study, a hydroalcoholic extract was obtained from the root of Valeriana pinnatifida R. & P., "valeriana," via maceration. Acute toxicity was determined at a limit dose of 2000 mg/kg using the OECD 423 protocol. The anticonvulsant effect was evaluated using an experimental model of pentylenetetrazol (PTZ)-induced seizures in mice. Five groups of six albino mice of the C53/CNPB strain were used: group 1 (saline control), group 2 (PTZ 80mg/kg), group 3 (diazepam 5mg/kg + PTZ), group 4 (EHVP 300mg/kg + PTZ), and group 5 (EHVP 400mg/kg + PTZ). The treatments were administered orally 30 minutes before the induction of seizures with PTZ. The results identified the secondary metabolites alkaloids and terpenes. The toxicity study showed a significant variation in the weight of the mice. The 300 mg/kg dose demonstrated a statistically significant anticonvulsant activity compared to the control group in terms of seizure severity and mortality. It is concluded that the hydroalcoholic root extract of Valeriana pinnatifida R. & P., "valeriana," at a concentration of 300 mg/kg, exhibits an anticonvulsant effect.Publicación Acceso abierto Actividad antiinflamatoria del extracto etanólico de las hojas del Baccharis buxifolia (Lam.) Pers. “Talla” leaves in mice. Solubility testing and a qualitative phytochemical profile of the ethanolic extract were carried out. To determine anti-inflammatory activity, the subplantar edema method with 1% albumin, as described by Winter et al. (modified), was employed. Forty-two albino mice (Mus musculus strain Balb/C53) of both sexes, each weighing more than 25 g, were used. The mice were randomly distributed into six groups and subsequently administered 0.1 mL of 1% albumin in the subplantar region. The plant extract was tested at concentrations of 100, 200, and 400 mg/kg of body weight, while dexamethasone (4 mg/kg) and diclofenac (50 mg/kg) were used as standard drugs. The results showed that the ethanolic extract of Baccharis buxifolia leaves, at 400 mg/kg, exhibited the greatest anti-inflammatory effect at two hours, with an inhibition percentage of 62%, compared with the lower extract concentrations (100 mg/kg: 10%; 200 mg/kg: 28%) and with the standard drugs diclofenac (50 mg/kg: 31%) and dexamethasone (4 mg/kg: 48%). It is concluded that the ethanolic extract of Baccharis buxifolia (Lam.) Pers. “Talla” leaves demonstrates significant anti-inflammatory activity at a dose of 400 mg/kg.Publicación Acceso abierto Actividad antimicótica "in vitro" del aceite esencial del fruto maduro de Schinus molle L. (Molle) against the Candida albicans ATCC 10231 strain. Methodology: The methodology used was experimental, explanatory, quantitative, and applied. The fruit was collected in the Huamanga province (Ayacucho). The essential oil (EO) was extracted by steam distillation. The antifungal activity was evaluated using the Kirby-Bauer method (disk diffusion). The strain was seeded on 18 plates with Sabouraud Dextrose Agar at a turbidity of 0.5 McFarland. The EO was evaluated at concentrations of 40%, 60%, 80%, and 100%, using Fluconazole 25 µg as a positive control and dimethyl sulfoxide as a negative control. The inhibition zones were measured with a digital vernier caliper at 24, 48, and 72 hours. Results: The EO of Schinus molle L. showed inhibition halos of 6 mm at 40 and 60%, 9.7 mm at 80%, and 12.3 mm at 100%. It is concluded that the EO at 40 and 60% presented no activity, but at 80 and 100% it presented antifungal activity against the Candida albicans ATCC 10231 strain.Publicación Acceso abierto Actividad antimicrobiana del aceite esencial de Origanum vulgare L. “orégano peruano” frente a Staphylococcus aureus(Universidad Privada Norbert Wiener, 2018-05-31) Salinas Segura, Roger; Carhuapoma Yance, MarioThe objective of this research was to confirm the antimicrobial activity of the essential oil of Origanum vulgare L., "Peruvian Oregano," against Staphylococcus aureus. The essential oil was obtained by steam distillation and purified with anhydrous sodium sulfate. The analysis of the main phytochemical components of the oil was performed using organoleptic and instrumental methods. The antimicrobial activity was assayed against Staphylococcus aureus ATCC 6538 using the disk diffusion method (Kirby-Bauer), with Gentamicin 10 µg (OXOID brand) as the positive control and dimethyl sulfoxide as the negative control. Results: The essential oil had a good yield of 2.02%, distilled in 60 minutes, with a density of 0.9132 g/mL and a refractive index of 1.475. The main components were carvacrol and thymol. The essential oil of Origanum vulgare L. showed antimicrobial activity against Staphylococcus aureus ATCC 6538, with inhibition halos of 11.47,13.40,16.60, and 19.65 mm at concentrations of 25,50,75, and 100 µg/mL, respectively. The gentamicin control resulted in an inhibition halo of 30.14 mm. Conclusion: The highest antimicrobial activity of the essential oil of Origanum vulgare L. "Peruvian Oregano" against Staphylococcus aureus was confirmed at 100 µg/mL.Publicación Acceso abierto Actividad antimicrobiana del extracto etanólico de las hojas de Schinus molle L. Schinus molle L. ("Molle"), a native tree with medicinal applications whose ethanolic extract exhibits antibacterial properties, is a clear example. The objective of this research was to confirm the antimicrobial activity of the ethanolic extract from the leaves of Schinus molle L. To this end, the ethanolic extract was obtained at concentrations of 500 and 1000 mg/mL, with 0.12% chlorhexidine gluconate as the positive control and distilled water as the negative control. The study of antimicrobial activity was conducted using the disk diffusion method with surface plating technique. The experimental solutions were incubated under anaerobic conditions for 48 and 72 hours at 37∘ C, after which the diameters of the inhibition halos were read with a vernier caliper. A qualitative analysis determined the presence of metabolites: flavonoids, alkaloids, carbohydrates, steroids and/or triterpenes, reducing sugars, and phenolic compounds. Results: The ethanolic extract at concentrations of 500 and 1000 mg/mL demonstrated antimicrobial activity against Streptococcus mutans ATCC 25175. From the comparison performed, chlorhexidine gluconate produced greater inhibition. Conclusion: The ethanolic extract of Schinus molle L. 'Molle' presents antimicrobial activity against Streptococcus mutans ATCC 25175, and the chlorhexidine gluconate is qualitatively similar to the ethanolic extract of Schinus molle L. This produces agro-industrial byproducts that should be utilized as an alternative for the root. Objective: To compare the antioxidant activity of the ethanolic extract from the root and fruit of Euterpe precatoria Mart., 'huasaí,' from the province of Maynas, relative to the species native to Chanchamayo. Methodology: The research design was quasi-experimental. The content of total phenols was determined by the Folin-Ciocalteu method, and antioxidant activity was determined by the 2,2-diphenyl-1-picrylhydrazyl (DPPH) method. Results: For antioxidant activity, linear regressions were used to obtain values for the half maximal inhibitory concentration (IC50). The 'huasaí' root from Chanchamayo and Maynas obtained an IC50 of 3.28 µg/mL and 3.86 µg/mL, respectively. The antioxidant activity measurements were contrasted using analysis of variance, Tukey's test (p<0.05), and the Student's t-test. Significant differences were found at the 5% level between the groups from the two provinces and for the three samples. The objective of this research was to evaluate the antioxidant activity of the ethanolic extract of the mesocarp of Hylocereus undatus (“pitahaya”) fruit and to determine its phytoconstituents. This quasi-experimental, analytical, descriptive, and prospective study prepared the ethanolic extract and evaluated its antioxidant activity using the DPPH method (2,2-diphenyl-1-picrylhydrazyl, Brand-Williams W). The antioxidant activity was expressed as IC50, with a result of 1.331 μg/mL, indicating significant antioxidant potential. Phytochemical analysis identified carbohydrates, reducing sugars, flavonoids, phenolic compounds, steroids, and alkaloids. The objective of this study was to determine the antioxidant and anti-inflammatory activity of the ethanolic extract of Passiflora tripartita var. mollissima leaves. The ethanolic extract was obtained by maceration, after which solubility testing and phytochemical screening were conducted to identify secondary metabolites. Antioxidant activity was evaluated using the DPPH method, while anti-inflammatory activity was assessed through the paw edema test induced by a phlogistic agent (1% albumin). The results demonstrated antioxidant activity at the following concentrations: 125 µg/mL (79.55%), 93.75 µg/mL (60.99%), 62.5 µg/mL (41.93%), and 31.25 µg/mL (21.82%). Compared with the TROLOX standard, the extract also showed significant anti-inflammatory activity, with a dose of 600 mg/kg producing an effect from the first hour of administration (p = 0.000), which was comparable to that of dexamethasone at 4 mg. The objective was to evaluate the efficacy of the antimicrobial and antioxidant activity of Schkuhria Pinnata, 'Canchalagua,' in inhibiting bacterial growth and reducing free radicals. A systematic intervention review study was conducted. Eligibility criteria were initially established, followed by a search in the Renati, Alicia (Concytec), Scielo, PubMed, Elsevier, Lilacs, Virtual Health Library (VHL), and Google Scholar databases. Studies were selected using the PRISMA method. A total of 103 articles were obtained, from which 11 studies were selected for data extraction and bias assessment, described by the Joanna Briggs Institute (2014). Metabolites were identified and quantified in different parts of the plant using precipitation, coloration, foaming, Folin-Ciocalteu, and aluminum chloride colorimetric methods. For the evaluation of antioxidant activity, the DPPH and ABTS methods were used, and the extracts showed reduction of free radicals. For antimicrobial activity, agar diffusion and dilution methods were employed. The extracts presented inhibition against Propionibacterium acnes, Pseudomonas aeruginosa, Escherichia coli, Enterococcus faecalis, Staphylococcus aureus, Mycobacterium smegmatis, Malephora lutea, and Staphylococcus epidermidis. Metabolites responsible for the antioxidant and antimicrobial activity were identified, and Schkuhria Pinnata, 'Canchalagua,' demonstrated efficacy in reducing free radicals and inhibiting bacteria.
