The pharmacological study was conducted with the objective of determining the hepatoprotective effect of the hydroalcoholic extract of the leaves of Momordica charantia L. (“bitter gourd”) in mice with acute liver intoxication induced by paracetamol, and of identifying the different secondary metabolites present in the plant species through the evaluation of the qualitative phytochemical profile. Our design was experimental. For the phytochemical analysis, the leaves of Momordica charantia L. (“bitter gourd”) were collected. Subsequently, the hydroalcoholic extract was obtained by the filtration method. For the pharmacological study, the hydroalcoholic extract of the leaves of Momordica charantia L. was administered at doses of 200, 400, and 600 mg/kg in mice weighing approximately 28–32 g of body weight. The extract was administered orally for three consecutive days, and on the following day, hepatotoxicity was induced with 600 mg/kg of paracetamol. At the end, all animals were subjected to administration of pentobarbital 6.5% 0.1 mL; sacrifice was carried out by the narcosis method, the livers were isolated, and histopathological studies were performed. Results: In the phytochemical screening, the presence of chemical compounds such as alkaloids, phenolic compounds, flavonoids, and tannins was identified. In the histopathological analysis, evidence of cellular damage at the hepatic level was found in all study groups due to the hepatotoxic action of paracetamol. Conclusions: The plant species possesses secondary metabolites with biological activity. The hydroalcoholic extract of the leaves of Momordica charantia L. (“bitter gourd”), at doses of 200, 400, and 600 mg/kg, did not demonstrate hepatoprotective effect in mice with acute paracetamol-induced intoxication.
Resumen
El estudio farmacológico se realizó con el objetivo de determinar el efecto hepatoprotector del extracto hidroalcohólico de las hojas de Momordica charantia L “caigua amarga” en ratones con intoxicación aguda hepática inducida por paracetamol e identificar los diferentes metabolitos secundarios presentes en la especie vegetal mediante la evaluación del perfil cualitativo fitoquímico. Nuestro diseño fue experimental, para el análisis fitoquimico se recolectaron las hojas de Momordica charantia L “caigua amarga”. Posteriormente se obtuvo el extracto hidroalcohólico por el método de filtración. Para el estudio farmacológico se empleó el extracto hidroalcohólico de las hojas de Momordica charantia L a dosis de 200, 400 y 600 mg/kg en ratones aproximadamente 28-32 g de peso corporal. Se administraron vía oral por 3 días consecutivos y al día siguiente se indujo a hepatotoxicidad con 600 mg/kg de paracetamol. Al final todos los animales fueron sometidos a la administración con pentobarbital 6,5% 0,1 mL, se realizó el sacrificio por el método de narcosis, se aislaron los hígados y se realizaron los estudios histopatológicos. Resultados: En el resultado del tamizaje fitoquímico se identificó la presencia de compuestos químicos como alcaloides, compuestos fenólicos, flavonoides, taninos. En el análisis histopatológico se identificó presencia de daño celular a nivel hepático en todos los grupos de estudio por la acción hepatotóxica del paracetamol. Conclusiones: la especie vegetal posee metabolitos secundarios con actividad biológica. El extracto hidroalcohólico de las hojas de Momordica charantia L, “caigua amarga” a dosis de 200, 400 y 600 mg/kg, no demostró efecto hepatoprotector en ratones con intoxicación aguda por paracetamol.
